To apply an effect on only a (moving) region of the picture you have to create a clip with transparent parts.
The manual says:

you need to point to an MLT clip with alpha transparency. [...] there are many ways we could create such clips, using for example a threshold filter or rotoscoping.

I know how to rotoscope, but how can I turn a rotoscoping sequence into an MLT clip?
