If a clip is relocated Kdenlive will fail to open the project and there will be a popup for selecting new location of the clip.
If I need to change more I open the project file with a text editor and use text search & replace.
Note: Always keep a copy before doing manual modification on project file.

As bigboss97 said changing the files manually in the project file is the most efficient way if you are simply moving the project files. However, I can think of two other ways of dealing with it.
1- If you create a project archive, when you reopen the archive it will ask you where to expand it and it will automagically fix the links in the project file to the new location. This is cumbersome when moving around on the same computer, but works well if moving to another PC via flash drive or external HDD/SDD.

2- Create a symlink with the name of the old location pointing to the new location.
